Route

The R407 begins in Willowmore, Eastern Cape, at a junction with the N9 national route (Knysna Street). It heads west for 93 kilometres, crossing into the Western Cape, to reach a junction with the N12 national route at Klaarstroom. The R407 joins the N12 westwards for 7 kilometres before becoming its own road westwards while the N12 turns to the north.

The R407 continues westwards for 42 kilometres to meet the northern terminus of the R328 route. It then turns northwards and heads for 6 kilometres to pass through the town of Prince Albert before continuing north-west for another 43 kilometres to reach its end at a junction with the N1 national route at Prince Albert Road.
